    The synthesis of Recombinant S.pneumoniae serotype 4 ply protein began with the isolation of ply gene from the native genome. The next step was to amplify the ply gene by inserting the ply gene into an expression vector and then introducing the vector into the selected host cell, Baculovirus. Significant amounts of recombinant S.pneumoniae serotype 4 ply protein were produced by the host only when expression genes were added. For the purpose of protein purification, the vector contains N-terminal 10xHis tag & C-terminal Myc tag to the ply DNA sequence. The purity of this protein is 90%+ determined by SDS-PAGE.

    ply (also known as SP_1923) is a gene encoding a protein named pneumolysin (ply) in Streptococcus pneumoniae serotype 4. The protein encoded by this gene is also known as thiol-activated cytolysin and belongs to Thiol-activated cytolysin family. The molecular weight of this protein is 53 kDa composed of 471 amino acids. It has a multiple biological activity, including the ability to lyse and interfere with the function of cells and soluble molecules of the immune system.
