BTLA
BTLA,即B和T淋巴細(xì)胞弱化因子,別名BTNL9、BTLA。它是一種主要在T細(xì)胞、B細(xì)胞和NK細(xì)胞上表達(dá)的免疫檢查點(diǎn)分子,由人體細(xì)胞自身的基因編碼。BTLA的作用機(jī)制涉及與其特異性配體HVEM(皰疹病毒入侵介質(zhì))結(jié)合,這種結(jié)合能夠抑制T細(xì)胞和B細(xì)胞的活化、增殖以及細(xì)胞因子的生成。BTLA與HVEM的結(jié)合通過激活免疫受體酪氨酸的抑制基序(ITIM)和免疫球蛋白酪氨酸開關(guān)基序(ITSM),導(dǎo)致SHP-1和SHP-2的磷酸化和募集,從而抑制T細(xì)胞的增殖和IL-2的產(chǎn)生,產(chǎn)生免疫抑制效應(yīng) 。BTLA參與了多種生理和病理過程的調(diào)節(jié),包括腫瘤、炎癥性疾病、自身免疫性疾病等。在腫瘤微環(huán)境中,BTLA的激活是PD-1抗體的潛在耐藥機(jī)制之一,BTLA的表達(dá)與腫瘤的進(jìn)展密切相關(guān)。在炎癥性疾病中,BTLA對(duì)T細(xì)胞介導(dǎo)的炎癥具有負(fù)調(diào)節(jié)作用。此外,在自身免疫性疾病中,BTLA的表達(dá)水平與疾病的活動(dòng)性相關(guān)。因此,BTLA是免疫學(xué)研究和相關(guān)疾病治療中的一個(gè)潛在靶點(diǎn) 。

Activity: Measured by its binding ability in a functional ELISA. Immobilized Human BTLA(CSB-MP773799HU1)at 2 μg/mL can bind Anti-BTLA recombinant antibody , the EC50 is 11.56-13.00 ng/mL.

Untransfected Jurkat cells surface (green line) and transfected Human BTLA Jurkat stable cells surface (red line) were stained with anti-BTLA antibody (2μg/1*106cells), washed and then followed by APC-conjugated anti-Human IgG Fc antibody and analyzed with flow cytometry.
