CRTC2
CRTC2,全稱為CREB調(diào)節(jié)轉(zhuǎn)錄共激活因子2,是一種與血糖調(diào)節(jié)密切相關(guān)的蛋白。該蛋白在胰島素信號通路中發(fā)揮重要作用,能夠結(jié)合并激活CREB蛋白,進而促進下游基因的表達。CRTC2蛋白的活性受到多種因素的調(diào)控,包括細胞內(nèi)鈣離子濃度、AMPK信號通路等。在胰島素作用下,CRTC2蛋白的活性增強,從而促進糖異生相關(guān)基因的表達,增加血糖水平。CRTC2蛋白與糖尿病的發(fā)生發(fā)展密切相關(guān),其過度激活會導致胰島素抵抗、脂肪肝和肥胖等代謝性疾病。針對CRTC2蛋白的藥物研發(fā)已成為糖尿病治療的重要方向。研究人員發(fā)現(xiàn),阿匹地靈C等天然產(chǎn)物能夠抑制CRTC2蛋白與CREB蛋白的結(jié)合,從而降低糖異生相關(guān)基因的表達,達到降低血糖的效果。在此基礎(chǔ)上,研究人員進一步設(shè)計了靶向CRTC2蛋白的先導化合物A57,該化合物對CRTC2蛋白的抑制作用比阿匹地靈C強30倍,能夠有效降低糖尿病小鼠的血糖和血脂水平。CRTC2蛋白相關(guān)藥物的研發(fā)為糖尿病治療提供了新的靶點和策略。
