FPGS
FPGS,即葉酰聚谷氨酸合酶,別名Folylpoly-gamma-glutamatesynthetase, 是一種在人類細(xì)胞中表達(dá)的酶,對(duì)于葉酸穩(wěn)態(tài)和增殖細(xì)胞的存活至關(guān)重要。FPGS的作用機(jī)制是通過(guò)催化ATP依賴的谷氨酸部分添加到葉酸和葉酸衍生物中,從而在細(xì)胞溶質(zhì)和線粒體內(nèi)建立和維持葉酰聚谷氨酸濃度。這種酶的活性對(duì)于甲氨蝶呤(MTX)類藥物的細(xì)胞內(nèi)代謝過(guò)程非常關(guān)鍵,它催化形成MTX多聚谷氨酸,這是MTX發(fā)揮抗腫瘤作用的活性形式。FPGS的異常表達(dá)或活性降低可能導(dǎo)致MTX耐藥,在癌癥治療中,F(xiàn)PGS的表達(dá)和活性是影響藥物敏感性的重要因素。

Overlay Peak curve showing HepG2 cells stained with CSB-RA983477A0HU (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1μg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1μg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
