GFRA3
GDNF家族受體α3(GFRA3)是神經(jīng)營(yíng)養(yǎng)因子受體家族的重要成員,常被稱為GFRα3或GDNF受體α3亞型。該蛋白主要通過(guò)與特定配體結(jié)合,協(xié)助調(diào)控神經(jīng)細(xì)胞的生長(zhǎng)與存活,尤其在外周和中樞神經(jīng)系統(tǒng)的發(fā)育中具有關(guān)鍵作用。GFRA3功能異常被發(fā)現(xiàn)與多種疼痛綜合征密切相關(guān),例如神經(jīng)病理性疼痛和慢性炎癥痛,患者常表現(xiàn)為痛覺(jué)過(guò)敏或異常性疼痛。在腫瘤領(lǐng)域,GFRA3的過(guò)表達(dá)可能促進(jìn)神經(jīng)母細(xì)胞瘤、甲狀腺髓樣癌等惡性腫瘤的進(jìn)展,部分研究提示其通過(guò)增強(qiáng)腫瘤細(xì)胞的侵襲能力影響預(yù)后。
針對(duì)GFRA3的藥物開(kāi)發(fā)集中在疼痛治療領(lǐng)域,已有企業(yè)開(kāi)發(fā)出選擇性拮抗劑進(jìn)入臨床前研究,旨在阻斷異常疼痛信號(hào)的傳導(dǎo)。在腫瘤治療方面,基于該受體設(shè)計(jì)的抗體偶聯(lián)藥物正在探索中,試圖通過(guò)靶向遞送細(xì)胞毒性物質(zhì)抑制腫瘤生長(zhǎng)。部分已上市的多靶點(diǎn)激酶抑制劑(如卡博替尼)被發(fā)現(xiàn)對(duì)GFRA3相關(guān)通路存在間接調(diào)節(jié)作用,這為老藥新用提供了潛在方向。
