HAS1
透明質(zhì)酸合成酶1(Hyaluronan Synthase 1,簡稱HAS1)是合成透明質(zhì)酸的關(guān)鍵酶之一,也被稱為HA synthase或細胞外基質(zhì)調(diào)節(jié)因子。HAS1通過催化葡萄糖醛酸和N-乙酰葡萄糖胺的聚合,生成透明質(zhì)酸分子,這種黏多糖廣泛存在于皮膚、關(guān)節(jié)液和細胞間質(zhì)中,具有維持組織水合度、促進創(chuàng)傷修復(fù)等功能。HAS1的異常表達與多種病理過程密切相關(guān)。在腫瘤微環(huán)境中,其過度活躍可能加速癌細胞轉(zhuǎn)移,尤其在乳腺癌和前列腺癌中表現(xiàn)突出;而在類風濕性關(guān)節(jié)炎患者中,關(guān)節(jié)滑膜細胞的HAS1活性升高會導(dǎo)致透明質(zhì)酸碎片堆積,引發(fā)炎癥反應(yīng)。近年來針對該靶點的藥物開發(fā)集中在抑制劑領(lǐng)域,例如通過小分子化合物干擾酶活性,已有部分候選藥物進入Ⅱ期臨床試驗,用于治療纖維化疾病和特定實體瘤。此外,基于透明質(zhì)酸的皮膚填充劑和關(guān)節(jié)潤滑劑已在臨床廣泛應(yīng)用,但其長效性和穩(wěn)定性仍依賴對HAS1調(diào)控機制的深入研究。
