IL26
IL26蛋白全稱為白細胞介素26，屬于IL-10細胞因子家族成員，因其主要表達于活化的T細胞和自然殺傷細胞，早期曾被稱為"NK細胞源性可溶因子"。該蛋白通過與細胞表面由IL-20R1和IL-10R2組成的異源二聚體受體結合，觸發(fā)免疫細胞活化并促進炎性介質(zhì)釋放。研究發(fā)現(xiàn)其在黏膜免疫防御中具有雙重角色，既能通過招募中性粒細胞增強抗菌能力，又會過度刺激Th17細胞導致組織損傷。臨床證據(jù)顯示，IL26在銀屑病患者的皮損部位、克羅恩病腸道病灶以及肺結核患者的肺部肉芽腫中異常高表達，其水平與疾病活動度呈正相關。針對該靶點的藥物開發(fā)尚處探索階段，已有研究嘗試通過單克隆抗體阻斷其與受體結合，或利用反義寡核苷酸抑制其產(chǎn)生。近期動物實驗表明，抑制IL26可顯著緩解關節(jié)炎模型的關節(jié)腫脹，但人體試驗尚未開展。由于IL26在抗病毒免疫中的保護作用，相關治療策略需精準平衡其促炎與抗感染功能的雙向調(diào)節(jié)。
