ITK，即白細(xì)胞介素-2誘導(dǎo)型T細(xì)胞激酶，別名Emt、Tsk。它是由人類ITK基因編碼的蛋白質(zhì)，主要在T細(xì)胞中表達(dá)。ITK的作用機(jī)制涉及T細(xì)胞受體（TCR）激活后的信號傳導(dǎo)過程，其通過被招募到細(xì)胞膜上，靠近被激活的TCR受體，并在那里被LCK激酶磷酸化。磷酸化導(dǎo)致ITK自磷酸化和完全激活。一旦激活，ITK會磷酸化PLCG1，導(dǎo)致這種脂酶的激活和隨后對其底物的切割。進(jìn)而內(nèi)質(zhì)網(wǎng)釋放鈣離子到細(xì)胞質(zhì)中，激活T細(xì)胞的核因子（NFAT）轉(zhuǎn)移到細(xì)胞核中執(zhí)行其轉(zhuǎn)錄功能。ITK對調(diào)節(jié)適應(yīng)性免疫反應(yīng)至關(guān)重要，它調(diào)控傳統(tǒng)T細(xì)胞和非傳統(tǒng)NKT細(xì)胞的發(fā)育、功能和分化。ITK對Th2和Th17細(xì)胞的發(fā)育和效應(yīng)功能具有重要作用，缺乏ITK的小鼠被證明不易患哮喘。

IHC image of CSB-RA954682A0HU diluted at 1:100 and staining in paraffin-embedded human lymph node tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
