LRRC15，即富含亮氨酸重復(fù)序列的蛋白15”，別名LIB。它是一種I型跨膜蛋白，屬于LRRC超家族成員之一，由人體細(xì)胞自身的基因編碼。LRRC15在多種腫瘤微環(huán)境中的基質(zhì)成纖維細(xì)胞中表達(dá)，并且其表達(dá)受TGFβ調(diào)控。LRRC15的作用機(jī)制涉及到與腫瘤微環(huán)境中的成纖維細(xì)胞相互作用，影響腫瘤細(xì)胞的遷移和侵襲，以及可能通過Wnt/β-catenin信號(hào)通路促進(jìn)三陰性乳腺癌細(xì)胞的遷移和侵襲。LRRC15在生物學(xué)上的意義非常廣泛，它不僅作為腫瘤微環(huán)境中的間充質(zhì)標(biāo)志物，還在腫瘤發(fā)展、轉(zhuǎn)移以及對(duì)免疫療法的反應(yīng)中發(fā)揮重要作用。LRRC15的高表達(dá)與多種實(shí)體瘤類型中抗PD-L1療法反應(yīng)不良相關(guān)，使其成為癌癥治療的潛在靶點(diǎn)。例如，ABBV-085是一種針對(duì)LRRC15的抗體-藥物偶聯(lián)物(ADC)，已在臨床前研究中顯示出對(duì)LRRC15陽性腫瘤的療效。此外，LRRC15+成纖維細(xì)胞的耗竭可以解除對(duì)腫瘤浸潤性CD8+T細(xì)胞的直接抑制，增強(qiáng)其效應(yīng)功能，并提高對(duì)抗PDL1免疫檢查點(diǎn)阻斷的腫瘤消退。因此，LRRC15是腫瘤免疫微環(huán)境研究和癌癥治療中的一個(gè)重要靶點(diǎn)。

Immunofluorescence staining of SH-SY5Y cells with CSB-PA819484LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C.
