MGA
MGA,即MAX二聚化蛋白MGA,別名MAD5、MXD5。它在人體中廣泛表達(dá),尤其在卵巢、睪丸等組織中表達(dá)量較高。MGA是一種雙特異的轉(zhuǎn)錄因子,含有3042個(gè)氨基酸,參與細(xì)胞命運(yùn)規(guī)范和RNA聚合酶II對(duì)轉(zhuǎn)錄的正調(diào)控。MGA在維持胚胎干細(xì)胞的多能性以及胚胎發(fā)育早期起著重要作用,是多梳抑制復(fù)合物PRC1.6的一個(gè)組分,通過(guò)穩(wěn)定PRC1.6復(fù)合物來(lái)發(fā)揮作用,主要沉默原始內(nèi)胚層關(guān)鍵基因Gata4/6及Sox17,避免胚胎干細(xì)胞向原始內(nèi)胚層的轉(zhuǎn)變。MGA的功能異常與多種疾病相關(guān),包括腫瘤和發(fā)育異常。MGA對(duì)于早期胚胎發(fā)育和細(xì)胞命運(yùn)決定具有關(guān)鍵作用,其研究有助于深入理解胚胎早期細(xì)胞命運(yùn)決定的分子機(jī)制。
