NKX2-1
NKX2-1,又名TTF-1,是一種具有特定同源結(jié)構(gòu)域的轉(zhuǎn)錄因子,對(duì)于甲狀腺、肺和腹側(cè)前腦的形態(tài)發(fā)生和分化至關(guān)重要。其編碼的蛋白質(zhì)含有特定的氨基酸序列和保守結(jié)構(gòu)域,能夠與DNA或其他蛋白質(zhì)相互作用以調(diào)控基因表達(dá)。NKX2-1參與調(diào)節(jié)神經(jīng)元祖細(xì)胞的特性、介導(dǎo)神經(jīng)元亞型的特化,并引導(dǎo)神經(jīng)元的遷移,同時(shí)參與多個(gè)重要的神經(jīng)系統(tǒng)發(fā)育信號(hào)通路。此外,NKX2-1的突變和缺失與多種疾病相關(guān),包括良性遺傳性舞蹈病、先天性甲狀腺功能減退和新生兒呼吸窘迫等,并可能與甲狀腺癌和神經(jīng)管畸形等疾病的發(fā)生有關(guān)。因此,對(duì)NKX2-1的深入研究不僅有助于理解其生物學(xué)功能,也為相關(guān)疾病的預(yù)防和治療提供了新的視角。

IHC image of CSB-RA181189A0HU diluted at 1:100 and staining in paraffin-embedded human lung tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.

The cells were fixed with 70% Ethylalcohol (18h) and then incubated in 10% normal goat serum to block non-specific protein-protein interactions followedby the antibody (1μg/1*106 cells) for 1 h at 4℃.The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 30min at 4℃. Control antibody (green line) was Rabbit IgG (1μg/1*106 cells) used under the same conditions. Acquisition of >10,000 events was performed.
