P2RX7

P2RX7全稱為嘌呤能受體P2X配體門控離子通道7，也被稱為P2X7受體或P2X7R。P2RX7廣泛分布于免疫細(xì)胞、神經(jīng)膠質(zhì)細(xì)胞等組織，其功能與細(xì)胞外三磷酸腺苷（ATP）濃度變化密切相關(guān)。當(dāng)ATP與受體結(jié)合后，會觸發(fā)離子通道開放，介導(dǎo)鉀離子外流及鈣離子內(nèi)流，進(jìn)而激活下游炎癥反應(yīng)通路。這種特性使其在NLRP3炎癥小體激活、細(xì)胞焦亡等病理過程中發(fā)揮關(guān)鍵作用，并與細(xì)胞增殖、凋亡調(diào)控存在復(fù)雜關(guān)聯(lián)。
P2RX7異常表達(dá)與多種疾病密切相關(guān)，包括類風(fēng)濕性關(guān)節(jié)炎、克羅恩病等自身免疫性疾病，阿爾茨海默病、多發(fā)性硬化等神經(jīng)系統(tǒng)病變，以及乳腺癌、黑色素瘤等惡性腫瘤的進(jìn)展。近年來針對該受體的藥物開發(fā)取得突破，如選擇性拮抗劑JNJ-54175446已完成銀屑病二期臨床試驗，AZD9056在慢性咳嗽治療中顯示出潛力。基于P2RX7的激活特性，科研人員正在探索其在腫瘤免疫治療中的增效作用，為精準(zhǔn)醫(yī)療提供新方向。
