PTPN22，即蛋白酪氨酸磷酸酶非受體22型。別名PEST-Domain Phosphatase、PTPN8、PEP。PTPN22基因位于1號(hào)染色體上，表達(dá)產(chǎn)物為Protein Tyrosine Phosphatase Non-Receptor Type 22。PTPN22是一種淋巴細(xì)胞特異性的細(xì)胞內(nèi)磷酸酶，與分子適配器蛋白CBL相關(guān)，可能參與調(diào)節(jié)T細(xì)胞受體信號(hào)通路中的CBL功能。該基因的突變與多種自身免疫性疾病有關(guān)，如1型糖尿病、類(lèi)風(fēng)濕關(guān)節(jié)炎、系統(tǒng)性紅斑狼瘡和Graves病。PTPN22在調(diào)節(jié)T細(xì)胞信號(hào)傳導(dǎo)中起關(guān)鍵作用，通過(guò)去磷酸化T細(xì)胞信號(hào)通路中的關(guān)鍵激酶如Lck、ZAP70、Vav和EB1來(lái)抑制T細(xì)胞抗原受體（TCR）信號(hào)傳導(dǎo)。此外，PTPN22還在血小板功能和動(dòng)脈血栓形成中發(fā)揮負(fù)調(diào)控作用，PTPN22缺乏會(huì)顯著縮短尾部出血時(shí)間并加速動(dòng)脈血栓形成，而不會(huì)影響靜脈血栓形成和凝血因子VIII和IX。
