PTPRT
PTPR，即蛋白酪氨酸磷酸酶受體T型，別名R-PTP-T、RPTPrho、RPTP-rho。PTPRT是一種受體型蛋白酪氨酸磷酸酶，屬于免疫球蛋白超家族，具有細胞外區(qū)、跨膜區(qū)和兩個串聯(lián)的細胞內(nèi)催化域。PTPRT的作用機制涉及其磷酸酶活性，能夠去磷酸化特定靶蛋白的酪氨酸殘基，從而調(diào)節(jié)細胞信號傳導。同時，PTPRT缺失提高了STING Y240位點酪氨酸磷酸化水平，抑制了STING的泛素化降解，從而增強了抗PD-1治療的效果。PTPRT的表達和活性與多種疾病的發(fā)生發(fā)展有關(guān)，包括癌癥和神經(jīng)系統(tǒng)疾病。PTPRT在中樞神經(jīng)系統(tǒng)的信號轉(zhuǎn)導和細胞粘附中具有作用，且與結(jié)直腸癌和食管鱗狀細胞癌有關(guān)。PTPRT的突變在多種癌癥類型中普遍存在，且與免疫治療的反應(yīng)性相關(guān)，是潛力較大的新型生物標志物和治療靶點。
