SNCA

SNCA,全稱α-突觸核蛋白,是神經(jīng)元中的一種重要蛋白,常見別名包括α-synuclein、SYN、PARK1、NACP等。該蛋白主要參與突觸小泡運(yùn)輸和神經(jīng)遞質(zhì)釋放,通過與多巴胺轉(zhuǎn)運(yùn)蛋白結(jié)合,調(diào)節(jié)其活性,并在多巴胺神經(jīng)傳遞中發(fā)揮重要作用。SNCA蛋白在帕金森病中扮演關(guān)鍵角色,其異常聚集形成路易小體,導(dǎo)致神經(jīng)元功能受損和死亡。研究表明,SNCA蛋白的基因突變與帕金森病密切相關(guān),其病理性翻譯后修飾,如Ser129位點(diǎn)的磷酸化,會使其錯誤折疊成多聚體,進(jìn)一步聚集成原纖維和淀粉樣纖維,損害神經(jīng)元功能。
目前,針對SNCA蛋白的藥物研發(fā)主要集中在免疫療法和生物藥開發(fā)上。被動免疫療法方面,多種抗SNCA抗體藥物正處于臨床II期、I期及臨床前階段,如Cinpanemab / BIIB054和Prasinezumab等。這些藥物通過直接或間接降低SNCA蛋白水平,或調(diào)節(jié)炎癥反應(yīng)來緩解帕金森病癥狀。此外,生物藥開發(fā)方面,也有多款針對SNCA蛋白的單抗和小分子拮抗劑進(jìn)入臨床試驗階段,如羅氏與Prothena開發(fā)的單抗藥物prasinezumab。這些進(jìn)展為帕金森病的治療提供了新的希望。

Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-Mouse IgG labeled by HRP and visualized using 0.05% DAB.

Immunofluorescence staining of SH-SY5Y cell with CSB-RA021912MA1HU at 1:30, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Mouse IgG(H+L).

Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1μg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1μg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
