SSTR5
SSTR5，即生長(zhǎng)抑素受體5，別名生長(zhǎng)抑素受體亞型5、SS5-R、Somatostatin receptor type 5等。SSTR5是一種G蛋白偶聯(lián)受體(GPCRs)，主要在垂體中高度表達(dá)，參與調(diào)控多種激素的分泌，如促腎上腺皮質(zhì)激素、催乳素和生長(zhǎng)激素等。SSTR5的作用機(jī)制涉及到與其天然配體生長(zhǎng)抑素(SST)或合成激動(dòng)劑藥物如奧曲肽(octreotide)結(jié)合，進(jìn)而抑制腺苷酸環(huán)化酶(AC)的活性，通過(guò)抑制性G蛋白(Gi)介導(dǎo)，調(diào)控胞內(nèi)cAMP濃度，影響激素的分泌和腫瘤生長(zhǎng)等過(guò)程 。SSTR5在生物學(xué)上的意義主要體現(xiàn)在其作為多種內(nèi)分泌疾病和腫瘤治療的潛在靶點(diǎn)。SSTR5的激活可以減少膽囊收縮，增加膽固醇性膽結(jié)石的風(fēng)險(xiǎn)，因此，拮抗SSTR5可能會(huì)促進(jìn)膽囊排空，減少膽囊結(jié)石的形成 。此外，SSTR5的表達(dá)水平與某些癌癥的發(fā)病有關(guān)，例如研究發(fā)現(xiàn)SSTR5基因的變異與頭頸部癌、肺癌、前列腺癌和腎癌等癌癥的發(fā)病有關(guān) 。因此，SSTR5不僅在調(diào)控激素分泌和腫瘤生長(zhǎng)中發(fā)揮重要作用，也是藥物開(kāi)發(fā)和疾病治療中的潛在靶點(diǎn)。
