TACSTD2
TACSTD2，即腫瘤相關(guān)鈣信號轉(zhuǎn)導(dǎo)子2，別名TROP2、EGP-1、GA733-1、M1S1等，由人類TACSTD2基因編碼，該基因位于1號染色體的1p32.1區(qū)域。TACSTD2蛋白是一種細(xì)胞表面糖蛋白，屬于單次跨膜蛋白，由323個氨基酸組成，具有一個疏水性前導(dǎo)肽、一個細(xì)胞外結(jié)構(gòu)域、一個跨膜結(jié)構(gòu)域和一個胞質(zhì)尾部。
TACSTD2的作用機制涉及其胞質(zhì)尾部的高度保守的磷脂酰肌醇4,5-二磷酸(PIP2)結(jié)合序列，該序列在信號轉(zhuǎn)導(dǎo)中起重要作用。TACSTD2通過與PIP2的相互作用以及酪氨酸和絲氨酸磷酸化位點參與細(xì)胞信號傳導(dǎo)，影響細(xì)胞周期進程。
TACSTD2在多種惡性腫瘤中過表達，與腫瘤的發(fā)生、侵襲和轉(zhuǎn)移有關(guān)，是一種重要的癌基因。TACSTD2的表達程度與疾病的惡性程度相關(guān)，其過表達可促進腫瘤細(xì)胞生長、增殖和轉(zhuǎn)移，故此成為了開發(fā)抗體偶聯(lián)藥物的新靶點。
