TERT
TERT,即端粒酶反轉錄酶,是端粒酶的核心亞基,由多個結構域組成,包括端粒酶保守基序結構域、催化域和RNA結合域等。它負責在細胞分裂過程中,通過反轉錄機制將RNA模板轉錄成DNA,并添加到染色體末端的端粒DNA序列中,從而維持端粒長度,確保細胞的持續(xù)增殖能力。TERT的表達受到嚴格調控,但在腫瘤細胞中常異常上調,與腫瘤的發(fā)生和發(fā)展密切相關。除了維持端粒長度,TERT還參與DNA修復、細胞存活、防止凋亡、促進生長和增殖等過程,并展現(xiàn)出獨立于端粒的新生物學功能,如基因轉錄調控、血管新生和免疫炎癥等。這些發(fā)現(xiàn)不僅為理解TERT的生物學作用提供了新的視角,也為治療端粒酶相關疾病提供了重要的理論基礎和潛在的治療策略。
