產(chǎn)品評(píng)價(jià): 我用CSB-E04578m-IS檢測(cè)小鼠血清中干擾素-γ,第一次買(mǎi)華美的試劑盒,跟著說(shuō)明書(shū)一步一步進(jìn)行操作,最終做出來(lái)的結(jié)果并沒(méi)有得到想要的統(tǒng)計(jì)學(xué)差異,可是有了相應(yīng)的趨勢(shì),還需要進(jìn)一步去做來(lái)驗(yàn)證。說(shuō)一下對(duì)于這個(gè)試劑盒的一些感受吧。 1.說(shuō)明書(shū)寫(xiě)的特別詳細(xì),一步一步來(lái)就能做出來(lái),比較簡(jiǎn)單,但是試劑寫(xiě)的是英文名字,大家用之前一定要確定好哪個(gè)是哪個(gè),別著急用錯(cuò)了,這個(gè)在說(shuō)明書(shū)里也有對(duì)應(yīng)的。 2.生物素標(biāo)記抗體和辣根過(guò)氧化物酶標(biāo)記親和素比較少,大家加樣時(shí)候要特別注意,特別精確!最好一開(kāi)始先渦旋一下,把附壁的渦旋下來(lái),我加的時(shí)候做了48個(gè)樣本,兩個(gè)我感覺(jué)用了2/3了,下次用時(shí)候估計(jì)得聯(lián)系這邊客服看能不能補(bǔ)加了 3.最后的結(jié)果里的標(biāo)準(zhǔn)曲線很不錯(cuò),做出來(lái)R方大于0.99,但是我組內(nèi)差異做的有一些大,可能和洗板操作有關(guān)?得最近再去試一下,但是我六個(gè)組整體的趨勢(shì)確實(shí)是能看出來(lái)了 4.官網(wǎng)的一些資料確實(shí)很不錯(cuò),那個(gè)標(biāo)曲處理的軟件也很專業(yè),但是就是可能得一個(gè)一個(gè)輸入x值得到y(tǒng)值,這個(gè)比較頭大。大家做之前一定要多看看視頻和說(shuō)明,不要悶頭直接做?。∶幻♂対舛?,同時(shí)做一下預(yù)實(shí)驗(yàn) 希望我接下來(lái)能出好的結(jié)果!也希望大家也能有好的實(shí)驗(yàn)結(jié)果??!科研人加油!

功能:Type II interferon produced by immune cells such as T-cells and NK cells that plays crucial roles in antimicrobial, antiviral, and antitumor responses by activating effector immune cells and enhancing antigen presentation. Primarily signals through the JAK-STAT pathway after interaction with its receptor IFNGR1 to affect gene regulation. Upon IFNG binding, IFNGR1 intracellular domain opens out to allow association of downstream signaling components JAK2, JAK1 and STAT1, leading to STAT1 activation, nuclear translocation and transcription of IFNG-regulated genes. Many of the induced genes are transcription factors such as IRF1 that are able to further drive regulation of a next wave of transcription. Plays a role in class I antigen presentation pathway by inducing a replacement of catalytic proteasome subunits with immunoproteasome subunits. In turn, increases the quantity, quality, and repertoire of peptides for class I MHC loading. Increases the efficiency of peptide generation also by inducing the expression of activator PA28 that associates with the proteasome and alters its proteolytic cleavage preference. Up-regulates as well MHC II complexes on the cell surface by promoting expression of several key molecules such as cathepsins B/CTSB, H/CTSH, and L/CTSL. Participates in the regulation of hematopoietic stem cells during development and under homeostatic conditions by affecting their development, quiescence, and differentiation.
