IL3，即白細(xì)胞介素3，別名Interleukin-3、MCGF。IL-3主要由活化的T細(xì)胞和NK細(xì)胞產(chǎn)生，此外，IL-1激活的人內(nèi)皮細(xì)胞、IgE激活的小鼠肥大細(xì)胞和胎盤(pán)細(xì)胞等也能產(chǎn)生IL-3。IL-3的作用機(jī)制主要通過(guò)與其受體結(jié)合并觸發(fā)細(xì)胞內(nèi)信號(hào)傳導(dǎo)過(guò)程來(lái)實(shí)現(xiàn)。IL-3與其受體α鏈（IL-3Rα）結(jié)合后，會(huì)進(jìn)一步與β鏈（IL-3Rβ，CD131）結(jié)合，形成異源二聚體或寡聚體，從而激活受體相關(guān)的Janus激酶（JAK），主要是JAK2。激活的JAK2募集STAT5并介導(dǎo)其磷酸化，磷酸化的STAT5進(jìn)入細(xì)胞核，誘導(dǎo)下游效應(yīng)基因的轉(zhuǎn)錄。此外，IL-3還激活Ras-MAPK和磷酸肌醇3-激酶（PI3K）通路。IL-3是一種重要的造血生長(zhǎng)因子，對(duì)多種細(xì)胞類型的發(fā)育起著關(guān)鍵作用，包括粒細(xì)胞、單核細(xì)胞、巨噬細(xì)胞、嗜酸性粒細(xì)胞、嗜堿性粒細(xì)胞和肥大細(xì)胞。IL-3還參與免疫調(diào)節(jié)、炎癥反應(yīng)以及血液系統(tǒng)疾病的治療。
