KIR2DL1
KIR2DL1是殺傷細(xì)胞免疫球蛋白樣受體家族的重要成員,全稱為殺傷細(xì)胞免疫球蛋白樣受體2DL1,也被稱作CD158a。KIR2DL1主要表達(dá)于自然殺傷細(xì)胞(NK細(xì)胞)表面,通過識別特定的人類白細(xì)胞抗原(HLA-C2亞型),向NK細(xì)胞傳遞抑制性信號,維持機(jī)體對健康細(xì)胞的免疫耐受。當(dāng)靶細(xì)胞表面HLA-I類分子表達(dá)異常時(shí),KIR2DL1的抑制作用減弱,NK細(xì)胞可被激活并殺傷靶細(xì)胞,這一機(jī)制在抗病毒感染和腫瘤免疫監(jiān)視中起關(guān)鍵作用。KIR2DL1基因多態(tài)性與白血病、艾滋病病程進(jìn)展及妊娠期子癇前期等疾病密切相關(guān)。針對該受體的免疫療法成為研究熱點(diǎn),例如通過單克隆抗體阻斷KIR2DL1與其配體結(jié)合,解除NK細(xì)胞活性抑制,已在多發(fā)性骨髓瘤和急性髓系白血病的臨床試驗(yàn)中顯示出治療潛力。部分藥物如IPH2101已完成II期試驗(yàn),為腫瘤免疫治療提供了新策略。

IHC image of CSB-RA012352MA1HU diluted at 1:100 and staining in paraffin-embedded human lung tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
