THPO
THPO,全稱促血小板生成素,是一種由人體內(nèi)THPO基因編碼的糖蛋白激素,也被稱為巨核細胞生長衍生因子(MGDF)。它主要由腎臟和肝臟分泌,能夠刺激巨核細胞的形成和分化,進而促進血小板的生成。THPO通過與MPL受體結(jié)合,激活下游信號通路,促進巨核細胞增殖、分化和成熟,最終形成血小板。在臨床上,THPO主要用于治療血小板減少癥,如原發(fā)免疫性血小板減少癥(ITP)和化療后血小板恢復,以及預防腫瘤治療引起的血小板減少。目前,全球已有多種THPO類似物和受體激動劑獲批上市,如重組人血小板生成素(rhTPO)、羅普司亭、艾曲泊帕等,這些藥物通過不同方式激活THPO受體,提高血小板計數(shù),改善患者的出血風險。此外,THPO在肝病和移植領域也有應用,如肝衰竭時TPO水平下降可作為預后標志物,而在干細胞移植中,TPO預處理可增強造血干細胞歸巢效率。目前,科學家們還在研究THPO在腫瘤治療副作用、骨髓纖維化等疾病中的應用,以及開發(fā)新型THPO類似物和受體激動劑,以進一步提高療效和安全性。
