TSLP

TSLP，全稱胸腺基質(zhì)淋巴細(xì)胞生成素，是一種由上皮細(xì)胞等多種細(xì)胞分泌的細(xì)胞因子，在免疫調(diào)節(jié)中扮演重要角色。它主要由上皮細(xì)胞、氣道平滑肌細(xì)胞、角質(zhì)形成細(xì)胞等分泌，能夠分化2型細(xì)胞中幼稚的CD4+T淋巴細(xì)胞，產(chǎn)生IL-4、IL-5、IL-13，并降低與1型細(xì)胞相關(guān)的γ干擾素（IFN-γ）的表達(dá)，對(duì)多種細(xì)胞產(chǎn)生影響。TSLP的異常表達(dá)與多種免疫相關(guān)疾病的發(fā)生和發(fā)展密切相關(guān)，如過(guò)敏性哮喘、特應(yīng)性皮炎和過(guò)敏性鼻炎等過(guò)敏性疾病，以及類風(fēng)濕性關(guān)節(jié)炎和系統(tǒng)性紅斑狼瘡等自身免疫性疾病。TSLP被證實(shí)與多種過(guò)敏性疾病有關(guān)，包括特應(yīng)性皮炎、支氣管哮喘、嗜酸性粒細(xì)胞食管炎等。此外，TSLP還與慢性炎癥性疾病，如慢性阻塞性肺病和乳糜瀉，以及牛皮癬、類風(fēng)濕性關(guān)節(jié)炎等自身免疫疾病有關(guān)。
TSLP靶向藥不僅具有廣譜的特點(diǎn)，能夠有效針對(duì)多個(gè)適應(yīng)癥，而且其針對(duì)的適應(yīng)癥還具備影響廣、病程長(zhǎng)的特點(diǎn)，具有極高的商業(yè)價(jià)值。在適應(yīng)癥上以治療哮喘居多，但是隨著深入的研究，在乳腺癌、白血病治療等方面也顯示靶點(diǎn)優(yōu)勢(shì)。

Measured by its binding ability in a functional ELISA. Immobilized human TSLP(R127A,R130A) at 2 μg/ml can bind Anti-TSLP recombinant antibody(CSB-RA025141MA2HU). The EC₅₀ is 52.55-58.67 ng/mL.

IHC image of CSB-RA025141MA1HU diluted at 1：100 and staining in paraffin-embedded human liver tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
